Resazurin Fluorescent Assay

The resazurin fluorescent assay is a cell-based method for estimating viability and metabolic activity, making it useful for evaluating neuronal health in neuroscience research. Living cells reduce the nonfluorescent dye resazurin to resorufin through cellular redox activity, producing a fluorescent signal that generally reflects the number and metabolic state of viable cells. Researchers apply the assay to cultured neurons and other neural cells to assess neurotoxicity, compare disease-related or experimental conditions, and evaluate potential neuroprotective compounds. Its sensitivity and compatibility with multiwell formats support quantitative analysis and higher-throughput screening, although results can also reflect changes in metabolism rather than cell number alone.

Most bacterial infections produce a biofilm. By virtue of their environment, biofilm associated bacteria are often phenotypically drug resistant. Novel antibacterial molecules that kill bacteria in biofilms are thus a high priority. We establish an assay to quickly screen for antimicrobial compounds that are effective at eradicating biofilms.

The charcoal agar resazurin assay (CARA) is a semi-quantitative, medium-throughput method to assess activity of test agents against mycobacteria that are replicating, non-replicating, or both. The CARA permits rapid evaluation of time- and concentration-dependent activity and identifies parameters to pursue by colony forming unit (CFU) assays.
