Rodent Stroke Model

Rodent stroke models are experimental systems that reproduce key features of human cerebrovascular injury in rats or mice, enabling controlled study of ischemic and hemorrhagic stroke. In a common ischemic model, researchers transiently or permanently block the middle cerebral artery, reducing regional blood flow and, when the obstruction is removed, producing reperfusion injury; tissue damage is then assessed through neurological testing, imaging, and histology. These models help clarify how interrupted circulation causes neuronal death, inflammation, blood-brain barrier disruption, and functional impairment. They also support evaluation of neuroprotective treatments, rehabilitation strategies, and mechanisms of recovery while revealing how experimental findings may translate to clinical neuroscience.

Various murine models of middle cerebral artery occlusion (MCAO) are widely used in experimental brain research. Here, we demonstrate the model of transcranial permanent distal MCAO which produces consistent cortical infarction of a size corresponding to damage imposed by the majority of human ischemic strokes.

The following report describes how to replicate the rodent model of nonarteritic anterior ischemic optic neuropathy (rNAION), using the appropriate dye, contact lens and laser parameters. We also reveal the appropriate steps for evaluating the rNAION lesion in vivo.

To simulate thromboembolic stroke, emboli prepared from heterologous rat blood were injected into the middle cerebral artery, followed by administration of systemic thrombolysis for recanalization. This version of the model is optimized for use in a multi-laboratory network and supports testing multiple candidate therapeutics.
