Spatiotemporal Neural Activity

Spatiotemporal neural activity describes how patterns of neuronal signaling are distributed across the nervous system and change over time. It arises as neurons generate electrical impulses and chemical signals that propagate through connected circuits, producing coordinated activity across different locations and timescales. Researchers investigate these dynamics using methods such as electrophysiology, calcium imaging, and functional neuroimaging to relate neural patterns to sensation, movement, learning, and behavior. Characterizing spatiotemporal activity helps reveal how neural circuits process information, synchronize responses, and adapt during development, disease, or changing environmental conditions.

We have developed an in vitro unfolded hippocampus which preserves CA1-CA3 array of neurons. Combined with the penetrating micro-electrode array, neural activity can be monitored in both the longitudinal and transverse orientations. This method provides advantages over hippocampal slice preparations as the propagation in the entire hippocampus can be recorded simultaneously.

This protocol describes the application of an engineered blue-light-activated allosteric switch (LightR) domain for reversible spatiotemporal control of protein activity. Utilizing Src tyrosine kinase as a model, this study offers an elaborate protocol for developing and characterizing light-regulated Src (LightR-Src). It demonstrates the versatility of this approach across enzyme classes.

Recently available video recording and spatiotemporal mapping (STmap) techniques make it possible to visualize and quantify both propagating and mixing patterns of intestinal motility. The goal of this protocol is to explain the generation and analysis of STmaps using the GastroIntestinal Motility Monitoring (GIMM) system.
