Superior Temporal Sulcus

The superior temporal sulcus (STS) is a cortical groove in the temporal lobe that separates the superior and middle temporal gyri and supports perception and social cognition. It integrates auditory, visual, and language-related information, with neural activity often responding to dynamic cues such as speech, biological motion, facial expressions, and shifts in gaze. Neuroscience research uses functional imaging, electrophysiology, and lesion studies to examine how the STS contributes to interpreting other people’s actions, intentions, and communication. Studying this region clarifies the organization of multisensory processing and helps characterize changes associated with developmental, psychiatric, and neurological conditions.

Here, we present a standardized series of protocols to observe the superior ocular sulcus, a recently-identified, evolutionarily-conserved structure in the vertebrate eye. Using zebrafish larvae, we demonstrate techniques necessary to identify factors that contribute to the formation and closure of the superior ocular sulcus.

The present work provides a comprehensive set of guidelines for manually tracing the medial temporal lobe (MTL) structures. This protocol can be applied to research involving structural and/or combined structural-functional magnetic resonance imaging (MRI) investigations of the MTL, in both healthy and clinical groups.
