Synaptosome Assay

A synaptosome assay is an experimental method that uses isolated, sealed nerve-terminal particles to investigate synaptic structure and function in neuroscience. Researchers typically prepare synaptosomes by homogenizing brain tissue and separating nerve-terminal fractions through differential centrifugation; these particles retain components such as plasma membranes, synaptic vesicles, mitochondria, and neurotransmitter transporters. Measuring neurotransmitter uptake or release, receptor activity, ion responses, and related biochemical signals reveals how synapses respond to drugs, toxins, genetic changes, or disease-associated conditions. Because synaptosomes preserve key presynaptic mechanisms while enabling controlled experiments, they support studies of neuronal communication, neuropharmacology, neurotoxicity, and synaptic dysfunction.

This protocol presents an in vitro live-imaging phagocytosis assay to measure the phagocytic capacity of astrocytes. Purified rat astrocytes and microglia are used along with pH indicator-conjugated synaptosomes. This method can detect real-time engulfment and degradation kinetics and provides a suitable screening platform to identify factors modulating astrocyte phagocytosis.
