Thrombolysis Studies

Thrombolysis studies investigate how medications dissolve blood clots and restore blocked blood flow, a central approach in research on acute ischemic stroke and other vascular disorders. These treatments typically use tissue plasminogen activator or related agents to convert plasminogen into plasmin, which breaks down fibrin within a thrombus and may reopen an occluded vessel. Neuroscience researchers evaluate treatment timing, recanalization, neural tissue survival, functional recovery, and the risk of bleeding. Such studies guide therapeutic protocols, improve patient selection, and support the development of safer clot-dissolving strategies for cerebrovascular disease.

To simulate thromboembolic stroke, emboli prepared from heterologous rat blood were injected into the middle cerebral artery, followed by administration of systemic thrombolysis for recanalization. This version of the model is optimized for use in a multi-laboratory network and supports testing multiple candidate therapeutics.

In-vitro thrombolysis assays have often struggled to replicate in-vivo conditions whether in the model thrombus being digested or in the environment in which thrombolysis is occurring. Herein, we explore how coupling the Chandler loop and Real-Time Fluorometric Flowing Fibrinolysis Assay (RT-FluFF) is used for high-fidelity, ex-vivo, clot lysis monitoring.
