Vascular Manipulation

Vascular manipulation is the deliberate alteration of blood vessels or blood flow to investigate how the vascular system influences tissue function, including neural activity and brain health. In neuroscience, researchers modify vessel diameter, perfusion, or vascular signaling through controlled experimental interventions, then measure resulting changes in cerebral blood flow, oxygen delivery, blood-brain barrier function, or neuronal responses. These approaches help clarify neurovascular coupling, the coordinated relationship between active neurons and local blood supply, and reveal how vascular dysfunction contributes to conditions such as stroke and neurodegenerative disease. Vascular manipulation also supports studies of brain physiology, therapeutic targets, and strategies for improving drug delivery to neural tissue.

To date, thick tissue defects are typically reconstructed by applying autologous tissue flaps or engineered tissues. In this protocol, we present a new method for engineering vascularized tissue flap bearing an autologous pedicle, to serve as a substitute to autologous flaps.

This film demonstrated a delicate injection technique used to achieve optimal contrasting of the vascular trees and illustrate the potential benefits resulting from a detailed analysis of the resulting specimen using µCT and histological serial sections.
