White Matter Undercut

White matter undercut is a neuroscience lesion model in which the white matter beneath a selected region of cerebral cortex is surgically severed, separating that cortex from deeper brain structures while leaving much of the cortical tissue intact. This disconnection interrupts axonal pathways that normally carry signals between the cortex, thalamus, brainstem, and other subcortical regions, altering local network activity and communication. Researchers use the model to investigate cortical reorganization, epileptogenesis, seizure propagation, and functional recovery after traumatic or ischemic injury. Comparing neural activity and behavior before and after the lesion helps clarify how disrupted connectivity influences brain plasticity and neurological disease.

Here we present methodology for the production of a focal stroke in murine white matter by local injection of an irreversible endothelial nitric oxide synthase (eNOS) inhibitor (L-Nio). Presented are two stereotactic variations, retrograde neuronal tracing, and fresh tissue labeling and dissection that expand the potential applications of this technique.

Partially isolated cortex (“undercut”) is an efficient animal model of posttraumatic epileptogenesis. Here we demonstrate how to make a novel surgical device and use it to make more precise and consistent lesions to generate this model.

This video demonstrates the precise technique for inducing a white matter stroke in an anesthetized mouse by injecting a nitric oxide inhibitor at targeted brain coordinates within the white matter. This leads to blood vessel constriction and inhibition of blood supply, resulting in neuronal damage and a brain stroke.

DTI of the Visual Pathway - White Matter Tracts and Cerebral Lesions

Diffusion tensor imaging (DTI) was performed to try to depict the major parts of the visual pathway. The goal was to use an FDA approved standard commercial workstation which could be used for everyday routine in order to try to reduce postoperative damage of the visual pathway in the patients.
