Amphetamine Overdose

Amphetamine overdose is a toxic state caused by excessive exposure to amphetamine or related stimulants, producing excessive central and peripheral nervous system activation. Amphetamine enters presynaptic neurons, disrupts vesicular monoamine storage, and promotes the release and transporter-mediated reversal of dopamine and norepinephrine, intensifying sympathomimetic effects. This toxicity can lead to agitation, hyperthermia, rapid heart rate, hypertension, seizures, arrhythmias, and altered consciousness. In pharmacology and clinical toxicology, understanding these mechanisms supports recognition of poisoning, assessment of severity, and development of effective emergency management strategies, while informing safer prescribing, substance-use education, and research on stimulant-related harm.
