Anti-infective Agents

Anti-infective agents are drugs that prevent, control, or eliminate infections caused by microorganisms, making them central to pharmacology and the treatment of infectious disease. They act through selective toxicity, targeting pathogen-specific processes such as bacterial cell-wall synthesis, microbial protein or nucleic-acid production, fungal membrane integrity, or viral replication while minimizing harm to host cells. Their use includes antibacterial, antiviral, antifungal, and antiparasitic therapy, with drug selection guided by the causative organism, infection site, susceptibility, and patient factors. Studying these agents clarifies therapeutic mechanisms, adverse effects, drug interactions, and the causes and consequences of antimicrobial resistance.

An Assay for Quantifying Antibody-Mediated Phagocytosis of Parasite-Infected Erythrocytes

2025

This video demonstrates a flow cytometry-based phagocytosis assay involving labeled and opsonized Plasmodium falciparum-infected erythrocytes by monocytes. Flow cytometry identifies fluorescence in monocytes, confirming the successful identification of opsonized erythrocytes through phagocytosis.

An In Vitro Assay to Examine the Antibody-Dependent Enhancement of Viral Infection

2025

This video demonstrates an assay to measure antibody-dependent enhancement (ADE) of Dengue virus infection using Zika Virus Convalescent Serum. The assay uses Dengue reporter viral particles (RVPs) encoding a green fluorescent protein. Serum antibodies against the Zika Virus cross-react with RVPs to form a complex. The bound antibodies enhance RVP uptake by cells expressing the Fc receptor, which is detected using flow cytometry.

Detection of SARS-CoV-2 Neutralizing Antibodies using High-Throughput Fluorescent Imaging of Pseudovirus Infection

The protocol described here outlines a fast and effective method for measuring neutralizing antibodies against the SARS-CoV-2 spike protein by evaluating the ability of convalescent serum samples to inhibit infection by an enhanced green fluorescent protein-labeled vesicular stomatitis virus pseudotyped with spike glycoprotein.

Preparation and In Vitro Characterization of Dendrimer-based Contrast Agents for Magnetic Resonance Imaging

This protocol describes the preparation and characterization of a dendrimeric magnetic resonance imaging (MRI) contrast agent that carries cyclen-based macrocyclic chelates coordinating paramagnetic gadolinium ions. In a series of MRI experiments in vitro, this agent produced an amplified MRI signal when compared to the commercially available monomeric analogue.

Introducing Experimental Agents into the Mouse

2023

Many investigations performed in mice (Mus musculus) require the administration of an experimental agent to the animal. For example, it may be of interest to test the efficacy of a specific therapy, to induce a pathologic condition, or to administer anesthesia or palliative care. In order to ensure safe and efficient delivery, it is important to consider a variety of factors prior to the administration of the treatment. This video, which reviews agent administration in the mouse, begins by...
