Drug Metabolite Neutralization

Drug metabolite neutralization is the reduction or removal of a drug metabolite’s pharmacological activity or toxicity, a key process in pharmacology and drug safety. It commonly occurs through biotransformation, in which enzymes modify metabolites by oxidation, reduction, or hydrolysis and then attach polar groups through conjugation reactions such as glucuronidation or sulfation, promoting renal or biliary excretion. Understanding these pathways helps researchers predict drug duration, adverse effects, active or toxic metabolites, and interactions that alter metabolic enzyme activity. This knowledge supports safer dose selection, therapeutic monitoring, antidote development, and evaluation of individual differences in drug response.

We describe here the procedures for the extraction and purification of mRNA and metabolites from Drosophila heads. We are applying these techniques to better understand the cellular perturbations underlying neuronal degeneration. These methodologies can be easily scaled and adapted for other "omic" projects.
