Infliximab Tnf

Infliximab is a biologic drug that targets tumor necrosis factor alpha (TNF-α), a key inflammatory mediator involved in immune signaling and tissue damage. As a chimeric monoclonal antibody, it binds soluble and membrane-bound TNF-α, preventing the cytokine from activating TNF receptors and reducing downstream inflammatory responses. In pharmacology, infliximab illustrates how targeted immunotherapy can modify disease mechanisms rather than provide only symptomatic relief. It is used to treat conditions such as rheumatoid arthritis, inflammatory bowel disease, psoriasis, and ankylosing spondylitis, while its immunosuppressive effects require attention to infection risk, dosing, and patient monitoring.

Drugs for Treatment of Crohn's Disease in IBD Using Biologic Agents: Anti-TNF

2024

Tumor Necrosis Factor (TNF), a proinflammatory cytokine, contributes significantly to the inflammation seen in Crohn's disease. It exists as soluble TNF and membrane-bound TNF, with actions mediated through TNF receptors (TNFR). TNFR activation leads to the release of proinflammatory cytokines, T-cell activation, collagen production, and leukocyte migration, all contributing to inflammation in Crohn's disease. Anti-TNF monoclonal antibodies, namely infliximab (Remicade), adalimumab (Humira),...

Generating an Organ Culture Model to Simulate Early-Stage Intervertebral Disc Disease

2025

This video demonstrates the development of a pro-inflammatory and degenerative organ culture model for early-stage intervertebral disc (IVD) disease using TNF-α. It outlines the procedure for injecting TNF-α into the intervertebral disc and explains the mechanisms through which TNF-α induces inflammation and degradation of the IVD.

This study used a multi-well plate microfluidic system, significantly increasing throughput of cell rolling studies under physiologically relevant shear flow. Given the importance of cell rolling in the multi-step cell homing cascade and the importance of cell homing following systemic delivery of exogenous populations of cells in patients, this system offers potential as a screening platform to improve cell-based therapy.
