Lead Optimization

Lead optimization is the systematic refinement of a promising chemical compound into a safer, more effective drug candidate, making it a central process in pharmacology and drug discovery. Researchers iteratively modify the lead structure and analyze structure–activity relationships to improve target potency and selectivity while balancing absorption, distribution, metabolism, excretion, and toxicity properties. Pharmacological assays and computational or medicinal chemistry approaches help identify compounds with stronger desired effects and fewer off-target activities. This process connects molecular design with biological testing, guiding candidate selection, reducing development risks, and supporting the development of therapies with suitable efficacy, safety, and drug-like properties.

We present an ECG protocol that is technically easy, inexpensive, fast, and affordable in small mice, and can be performed with enhanced sensitivity. We suggest this method as a screening approach for studying pharmacological agents, genetic modifications, and disease models in mice.
