Rotational Trap Stiffness

Rotational trap stiffness is a measure of how strongly a rotational constraint resists angular displacement, typically expressed as restoring torque per unit angle. In a rotational optical trap, for example, light transfers angular momentum to a trapped particle, and small rotations generate a restoring torque that is approximately proportional to the angular displacement, allowing stiffness to be determined from torque or angular-fluctuation measurements. This concept helps quantify rotational stability, energy landscapes, and mechanical response in microscopic systems. Applications include optical tweezers, microrheology, torque measurements, and studies of molecular motors, colloids, and other rotationally driven processes.

Here, a method that enables quick, efficient, and inexpensive preparation of polyacrylamide gels in a multiwell plate format is described. The method does not require any specialized equipment and could be easily adopted by any research laboratory. It would be particularly useful in research focused on understanding stiffness-dependent cell responses.
