Measurement Invariance

Measurement invariance is a statistical property indicating that a measure assesses the same underlying construct in the same way across groups, settings, or time points. Researchers evaluate it by comparing increasingly constrained models, often through multigroup confirmatory factor analysis: configural invariance tests shared factor structure, metric invariance equates loadings, and scalar invariance equates intercepts, with stricter levels sometimes testing residual variances. Establishing appropriate invariance determines whether group differences reflect genuine differences in the construct rather than changes in how respondents interpret or answer items. It is therefore essential in survey research, longitudinal studies, psychometrics, and fair comparisons of latent means.

Isolating Invariant Natural Killer T Cells from a Mouse Model

This video illustrates a method for isolating invariant natural killer T cells, also known as iNKT cells. The procedure begins with injecting a synthetic glycolipid into a mouse to stimulate the activation and proliferation of iNKT cells in the spleen. Subsequently, single-cell suspensions are prepared from the spleen and incubated with iNKT-cell-recognizing complexes labeled with fluorophores, followed by magnetic bead-based separation, to isolate the iNKT cells.

Here we present an adapted protocol that can be used to generate a large number of murine invariant natural killer T cells from mouse spleen. The protocol outlines an approach by which splenic iNKT cells can be enriched for, isolated and expanded in vitro using a limited number of animals and reagents.

Optimized sampling protocols and the development of new wipe materials can be facilitated by standardized measurements of collection efficiency from wipe-sampling. Our approach for sampling trace explosives uses an automated device to control speed, force, and distance during wipe-sampling followed by extraction of collected explosives.
