Bir enzimin devir sayısı, birim zamanda dönüştürebileceği maksimum substrat molekülü sayısıdır. Çoğu enzimin devir sayısı saniyede 1 ila 1000 molekül arasında değişir. Katalaz, saniyede 2,8×106 molekül hidrojen peroksiti su ve oksijene dönüştürebilen, bilinen en yüksek devir sayısına sahiptir. Lizozim, saniyede yarım molekül ile bilinen en düşük devir sayısına sahiptir.

 Kimotripsin, sindirim sırasında proteinleri parçalayan pankreatik bir enzimdir. Kimotripsinin devir sayısı saniyede 100 moleküldür. Bu reaksiyon katalizlenmeden meydana gelseydi, peptid bağlarının nötr pH'taki suda parçalanması yüzlerce yıl alırdı.  Ribuloz 1,5-bisfosfat karboksilaz oksijenaz veya RuBisCO enzimi, saniyede 3 CO2 molekülünü sabitleyen çok düşük bir devir sayısına sahiptir ve en yavaş enzimlerden biridir. Ancak RuBisCO'nun doğadaki bolluğu, düşük ciro sayısını telafi eder. RuBisCO, yapraklarda bulunan toplam proteinin yaklaşık %50'sini oluşturur.

how fast enzymes work in digestion

Enzymes can work at very different speeds, and their turnover number shows how fast they convert substrate molecules into products. Turnover number is the maximum number of substrate molecules an enzyme can transform per unit time. For most enzymes, this value is between 1 and 1000 molecules per second.

Some enzymes are especially fast. Catalase has the highest known turnover number and can convert up to 2.8×10^6 molecules of hydrogen peroxide into water and oxygen each second. Lysozyme has the lowest known turnover number, at half a molecule per second.

Digestion also depends on enzyme speed. Chymotrypsin is a pancreatic enzyme that breaks down proteins during digestion, and its turnover number is 100 molecules per second. Without an enzyme, peptide bonds would take hundreds of years to break in water at neutral pH. Chymotrypsin helps make protein digestion in the intestine much faster.

Not every slow enzyme is unimportant. RuBisCO, or ribulose 1,5-bisphosphate carboxylase oxygenase, has a very low turnover number and fixes only 3 molecules of CO2 per second. It is one of the slowest enzymes, but it is very abundant in nature. RuBisCO makes up around 50% of the total protein found in leaves.

A high turnover number does not always mean high catalytic efficiency. Catalytic efficiency is the ratio of turnover number, kcat, to affinity, KM. An enzyme also needs a low KM for its substrate to be efficient. The average catalytic efficiency of most enzymes is about 10^5 M^-1 s^-1, so they are moderately efficient. A few enzymes reach 10^8 to 10^9 M^-1 s^-1 and are called superefficient or catalytically perfect.
