Protein Blot

Protein blotting is a laboratory method used to separate, detect, and analyze specific proteins within a complex biological sample, making it important for biochemical research. In a typical workflow, proteins are separated by gel electrophoresis, transferred to a membrane, and identified when primary antibodies bind their target proteins and labeled secondary antibodies generate a detectable signal. Researchers use protein blots to compare protein abundance, estimate molecular mass, assess post-translational changes, and validate findings from other experiments. The technique supports studies of cell signaling, gene expression, disease mechanisms, and protein purification by providing qualitative or semiquantitative molecular evidence.

This video demonstrates a quantitative dot blot, or QDB, technique for the absolute quantification of proteins of interest in a tissue sample. The proteins in the sample are immobilized on the nitrocellulose membrane inside the wells of a QDB plate. Utilizing target protein-specific antibodies labeled with a peroxidase enzyme, a chemiluminescent substrate is oxidized to produce light, which is measured to determine the protein concentration of the sample.

Western Blotting is used to identify the presence of specific proteins in electrophoretically separated samples. Following separation by a technique known as sodium dodecyl sulfate polyacrylamide gel electrophoresis, or SDS-PAGE, western transfer is used to move proteins from a polyacrylamide gel onto a piece of membrane which traps the proteins in their respective locations.
