2d Culture

2D culture is a laboratory method for growing cells as a monolayer on a flat, usually tissue-culture-treated surface under controlled conditions. Cells attach to the substrate, spread across its two-dimensional plane, and receive nutrients, gases, and signaling factors from a surrounding culture medium, allowing researchers to monitor proliferation, morphology, and cellular responses. In bioengineering, 2D culture supports cell expansion, biomaterial testing, disease modeling, and evaluation of drugs or engineered microenvironments. Although it does not reproduce the three-dimensional architecture of native tissue, its accessibility, reproducibility, and relatively low cost make it an essential foundation for developing and validating more complex culture systems.

This video describes the technique for long term 2D co-culture of leukemia cells with bone marrow stromal cells to study their interaction with the bone marrow microenvironment. The 2D coculture model utilizes the acute lymphoblastic leukemia cells and the bone marrow stromal or osteoblasts that influence tumor cell phenotype. This study will support investigation of the mechanisms that underlie bone marrow supported survival of leukemic cells during chemotherapy exposure.

Leukemic Subpopulation Harvest: A Method for Spatial Separation of Leukemic Cell Subpopulations from 2D Co-culture

This video describes a method for spatial separation of three leukemic cell subpopulations: the suspended leukemic cells freely floating in the media, phase bright leukemic cells adhered to the surface of the mesenchymal stromal cell monolayer, and the phase dim leukemic cells that have migrated beneath the mesenchymal stromal cell monolayer from 2D co-culture.
