Basal Membrane Extract

Basal membrane extract is a protein-rich extracellular matrix preparation used to recreate aspects of the basement membrane in cell culture, enabling cells to grow in a more tissue-like three-dimensional environment. It typically contains structural proteins such as laminin and type IV collagen, along with other matrix components, and can form a gel under physiological temperature conditions. Researchers use this matrix for organoid generation, cell differentiation, invasion assays, and tissue engineering, where matrix composition and stiffness influence cell adhesion, polarity, signaling, and morphology. By supporting controlled three-dimensional culture, basal membrane extract helps model tissue organization and evaluate biological responses in developmental, disease, and drug research.

Here we demonstrate our protocol for isolation of basal and submucosal gland duct cells from mouse tracheas. We also demonstrate the method of injecting stem cells into the dorsal mouse fat pad to create an in vivo model of submucosal gland regeneration.
