Target Protein Incubation

Target protein incubation is a biological technique in which a protein of interest is held under controlled conditions with selected reagents, cells, or other biomolecules to enable a measurable interaction or reaction. The process depends on factors such as temperature, pH, incubation time, protein concentration, and reagent compatibility, which influence molecular binding, enzymatic activity, folding, or modification. Researchers use target protein incubation in assays that characterize protein function, evaluate ligand or antibody binding, optimize purification and detection steps, and study biochemical pathways. Careful control of incubation conditions improves reproducibility and helps distinguish specific molecular effects from nonspecific interactions.

2025

In this video, we demonstrate the phage display technique to isolate pre-engineered phages expressing proteins with high affinity to the target protein.

Immunofluorescence Assay to Evaluate Effect of Target Proteins on Neurite Outgrowth

2025

In this video, we demonstrate an EGFP-based immunofluorescence assay to identify the efficacy of target proteins on the neurite outgrowth of primary cortical neurons. The primary neurons are transfected with a fluorescent protein, EGFP, and a target protein, and then an immunoassay is performed to investigate the effect of the target protein on neurite outgrowth.

Assessing the Internalization of Target Surface Proteins Using a Biotin Derivative

This video demonstrates a method to assess target protein internalization in the mouse cortical astrocytes using biotinylation, followed by cell lysis, streptavidin-based protein extraction, denaturation, and Western blot analysis to confirm successful internalization.

Quantitative Dot Blot to Estimate Target Proteins in a Tissue Lysate

2025

This video demonstrates a quantitative dot blot, or QDB, technique for the absolute quantification of proteins of interest in a tissue sample. The proteins in the sample are immobilized on the nitrocellulose membrane inside the wells of a QDB plate. Utilizing target protein-specific antibodies labeled with a peroxidase enzyme, a chemiluminescent substrate is oxidized to produce light, which is measured to determine the protein concentration of the sample.
