Hatching Protocol

A hatching protocol is a standardized procedure for supporting and monitoring the emergence of an embryo from an egg, cyst, or protective оболage under controlled biological conditions. It typically coordinates incubation conditions, timing, handling, and observation so that developmental progress can be assessed while minimizing stress or damage to the organism. In biology, hatching protocols help researchers obtain synchronized experimental organisms, measure developmental outcomes, and improve reproducibility across studies. They are used in developmental biology, ecology, aquaculture, and laboratory model systems to investigate embryogenesis, survival, behavior, and responses to environmental factors.

Here, we present a protocol to investigate the host-tissue distribution, transmission mode, and effect on the host fitness of a densovirus within a lepidopteran species, the cotton bollworm. This protocol can also be used for studying the interaction between other orally-transmitted viruses and their insect hosts.
