Hybridization Chain Reaction

Hybridization Chain Reaction (HCR) is an isothermal nucleic acid amplification method that converts recognition of a specific target sequence into a growing DNA or RNA polymer, enabling sensitive molecular detection without enzymatic amplification. In HCR, an initiator sequence opens one metastable hairpin, exposing a sequence that opens a second hairpin; alternating hybridization propagates a chain of linked hairpins under constant-temperature conditions. In cancer research, HCR can visualize and quantify RNA or DNA biomarkers in cells and tissues while preserving spatial information about tumor-associated gene expression. Its enzyme-free design supports multiplexed imaging and can improve analysis of heterogeneous tumors and biomarker networks.
