Capillary Electrophoresis-mass Spectrometry

Capillary electrophoresis-mass spectrometry (CE-MS) is an analytical technique that separates charged chemical species by electrophoretic mobility and identifies them according to their mass-to-charge ratio. During analysis, a high-voltage electric field drives ions through a narrow capillary, where differences in size, charge, and interactions with the surrounding electrolyte produce separation; emerging compounds are then ionized and transferred into a mass spectrometer for mass analysis. This coupled approach combines high separation efficiency with molecular identification and can characterize metabolites, peptides, proteins, pharmaceuticals, and other complex mixtures. In chemistry, CE-MS supports trace analysis, structural characterization, and studies of composition when sample amounts are limited or conventional chromatographic methods provide insufficient resolution.

A detailed protocol is described for the separation, identification, and characterization of proteoforms in protein samples using capillary zone electrophoresis-electrospray ionization-tandem mass spectrometry (CZE-ESI-MS/MS). The protocol can be used for the high-resolution characterization of proteoforms in simple protein samples and the large-scale identification of proteoforms in complex proteome samples.
