Fibronectin Film

Fibronectin film is a thin layer of the extracellular-matrix glycoprotein fibronectin deposited on a material surface, where it modifies surface chemistry and supports biological interactions. The film forms when fibronectin molecules adsorb to a substrate and may rearrange or unfold, exposing domains that bind cell-surface integrins and other matrix components; solution conditions and substrate properties influence its organization and stability. In chemistry and biomaterials research, fibronectin films functionalize otherwise inert surfaces for cell culture, tissue-engineering scaffolds, and biosensor studies. Their composition and molecular arrangement help researchers investigate protein adsorption, cell adhesion, and the design of bioactive interfaces.

Source: Logan G. Kiefer, Andrew R. Falkowski, and Taylor D. Sparks, Department of Materials Science and Engineering, The University of Utah, Salt Lake City, UT Electroplating is a process that uses electric current to reduce dissolved metal cations so that they form a thin coating on an electrode. Other thin film deposition techniques include chemical vapor deposition (CVD), spin coating, dip coating, and sputter deposition among others. CVD uses a gas-phase precursor of the element to be...

Fibronectins Connect Cells with ECM

2023

Fibronectin is an adhesive glycoprotein present in the extracellular matrix of embryogenic and adult tissue. These molecules primarily aid in regulating cell motility and attachment. A fibronectin molecule is composed of two identical polypeptide chains attached to each other by a pair of disulfide bonds at the C-terminal. Both proteoglycans and collagen are attached to fibronectin proteins, which, in turn, are attached to integrin proteins. These integrin proteins interact with transmembrane...

The goal of this protocol is to show the assembly of a biomimetic nanomatrix (NM) with Janus base nanotubes (JBNTs) and fibronectin (FN). When co-cultured with human mesenchymal stem cells (hMSCs), the NMs exhibit excellent bioactivity in encouraging hMSCs adhesion.

This method is useful for quantifying the early dynamics of cellular adhesion and spreading of anchorage-dependent cells onto the fibronectin. Furthermore, this assay can be used to investigate the effects of altered redox homeostasis on cell spreading and/or cell adhesion-related intracellular signaling pathways.
