Quantitative Nmr Integration

Quantitative NMR integration is a method for determining the relative or absolute amount of a compound by measuring the areas of its nuclear magnetic resonance signals. Because each signal’s integrated area is proportional to the number of contributing nuclei, accurate results require well-resolved peaks, a known reference or internal standard, and sufficient relaxation between scans to produce reliable signal intensities. In chemistry, quantitative NMR supports purity assessment, concentration measurements, reaction monitoring, and mixture analysis without requiring compound-specific calibration curves. Its broad applicability and minimal sample preparation make it a valuable complement to chromatographic and spectroscopic methods.

In this video, we demonstrate the in-cell nuclear magnetic resonance spectroscopy technique to study protein-ligand interactions between unlabeled overexpressed proteins and small molecules. The successful ligand-protein interaction is confirmed visually by the appearance of an additional set of peaks in the spectral region of interest that gradually replaces the original peaks.

A simple and non-destructive technique that measures the average content of drug substances in formulated drug products containing fluorine using low-field fluorine-19 (19F) time-domain (TD) nuclear magnetic resonance (NMR) is presented here. The technique can be applied to the development and manufacturing of drugs in the pharmaceutical industry.
