Air Accumulation

Air accumulation is the abnormal buildup of gas within a body cavity, organ, or tissue, and it matters clinically because it can disrupt normal anatomy and impair organ function. It may occur when air enters from outside the body, forms through injury or disease, or cannot be adequately absorbed or expelled; increasing gas volume can raise local pressure and compress nearby structures. Clinicians evaluate its location, amount, and cause using physical examination and imaging, then guide treatment according to severity. Recognizing air accumulation supports timely diagnosis of conditions such as pneumothorax and postoperative complications and helps prevent respiratory or circulatory compromise.

An Assay to Measure the Accumulation of a Fluorescent Antibiotic Probe in Bacterial Cells

This video demonstrates an assay to measure the accumulation of a fluorescent antibiotic probe in bacterial cells. Exposure to an efflux pump inhibitor disrupts the transmembrane proton gradient in bacterial cells, upon which a fluorophore-conjugated antibiotic is added. Inhibition of efflux pumps due to the disrupted proton gradient causes accumulation of the antibiotic inside the bacteria. Bacterial cells are lysed, and the fluorescence intensity is measured to confirm antibiotic accumulation.

The heterogeneous intra-tumoral accumulation of liposomes has been linked to an abnormal tumor microenvironment. Herein methods are presented to measure tumor microcirculation by perfusion imaging and elevated interstitial fluid pressure (IFP) using an image-guided robotic system. Measurements are compared to the intra-tumoral accumulation of liposomes, determined using volumetric micro-CT imaging.
