Ph Partitioning

pH partitioning is the distribution of a substance between aqueous and lipid phases as a function of the surrounding solution’s pH, a principle that helps explain how ionization influences biological behavior. Weak acids and bases shift between charged and uncharged forms as pH changes; the uncharged fraction generally crosses lipid membranes more readily, while the ionized fraction remains more soluble in water. In clinical science, this relationship supports predictions of drug absorption, membrane permeation, tissue distribution, and urinary excretion, and informs formulation and dosage decisions. Understanding pH partitioning also helps interpret how body compartments and physiological pH differences can alter medication response.

The distribution law or Nernst's distribution law is the law that governs the distribution of a solute between two immiscible solvents. This law, also known as the partition law, states that if a solute is added to the mixture of two immiscible solvents at a constant temperature, the solute is distributed between the two solvents in such a way that the ratio of solute concentrations in the solvents remains constant at equilibrium. For extracting a solute from an aqueous phase into an organic...

2012

A technique to probe the lipid raft partitioning of fluorescent proteins at the plasma membrane of living cells is described. It takes advantage of the disparity in diffusion times of proteins located inside or outside of lipid rafts. Acquisition can be performed dynamically in control conditions or after drug addition.

Here, we present a protocol to perform a quantitative analysis of the level of plasma-membrane association for fluorescently-tagged peripherally-associated protein. The method is based on the computational decomposition of membrane and cytoplasmic component of signal observed in cells labeled with plasma membrane fluorescent marker.

Metal-silicate Partitioning at High Pressure and Temperature: Experimental Methods and a Protocol to Suppress Highly Siderophile Element Inclusions

We present a procedure to determine the metal-silicate partitioning of siderophile elements, emphasizing techniques that suppress the formation of metal inclusions in experiments for the noble metals. The results of these experiments are used to demonstrate the effect of core-formation on the highly siderophile element composition of the mantle.
