Light-responsive Proteins

Light-responsive proteins are molecules that detect specific wavelengths of light and convert them into biochemical signals, making them valuable tools for studying how biological systems develop. Many contain a light-absorbing chromophore; photon absorption changes the protein’s conformation, which can alter its enzymatic activity, binding interactions, localization, or downstream signaling, often in a reversible and precisely timed manner. In developmental biology, these proteins help researchers control signaling pathways, gene expression, cell migration, and differentiation with spatial and temporal precision. Their use can reveal how localized cues and signaling dynamics shape tissue formation, while supporting the development of optogenetic approaches for manipulating living cells.

A Behavioral Assay to Measure Responsiveness of Zebrafish to Changes in Light Intensities

We developed the Visual-Motor Response to quantitate the motor output of larval zebrafish in response to light increments and decrements. We also examined zebrafish vision mutants, including the no optokinetic response (nrc) mutants, which were thought to be completely blind when tested by another vision assay, the optokinetic reflex.

Our overall aim is to understand how cells sense extracellular cues that lead to directed axonal growth. Here, we describe the methodology of Light-Induced Molecular Adsorption of Proteins, used to produce defined micro-patterns of extracellular matrix components in order to study specific events that govern axon outgrowth and pathfinding.
