Mammary Stem Cells

Mammary stem cells are self-renewing cells that generate the epithelial lineages forming and maintaining the mammary gland, making them central to tissue development and homeostasis. During mammary gland morphogenesis, they respond to local signals and systemic hormones that regulate proliferation, self-renewal, and differentiation into luminal and myoepithelial cells. Their activity supports ductal elongation, branching, and remodeling during puberty, pregnancy, and tissue repair. Studying mammary stem cells helps explain normal gland development, lineage organization, and how disrupted growth control may contribute to breast cancer, while also informing research on tissue regeneration and breast disease models.

A Novel Mammary Fat Pad Transplantation Technique to Visualize the Vessel Generation of Vascular Endothelial Stem Cells

This work demonstrates a novel approach to assess the proliferation, differentiation, and vessel-forming potential of vascular endothelial stem cells (VESCs) through mammary fat pad transplantation followed by whole-mount tissue preparation for microscopic observation. A lineage tracing strategy to investigate the behavior of VESCs in vivo is also presented.

This protocol describes methods for purifying, quantitating, and characterizing extracellular vesicles (EVs)/exosomes from non-adherent/mesenchymal mammary epithelial cells and for using them to transfer mammary gland-forming ability to luminal mammary epithelial cells. EVs/exosomes derived from stem-like mammary epithelial cells can transfer this cell property to cells that ingest the EVs/exosomes.

This protocol details the derivation of transplantable hematopoietic stem cells from mouse embryonic stem cells (ESC) and their subsequent injection into lethally irradiated recipient mice. Briefly, ESC are differentiated as embryoid bodies, which are then infected with retroviral HoxB4 and co-cultured with OP9 stromal cells and hematopoietic cytokines.

In this report, we demonstrate a system to isolate and culture donor cells from the mouse mammary gland, and orthotopically transplant these cells in recipient mice to analyze stromal: epithelial interactions during mammary tumor development.
