Optic Lobe Imaging

Optic lobe imaging is the use of microscopy to visualize the organization, growth, and cellular changes of the optic lobe, a neural structure involved in visual processing. By labeling cells or tissues with fluorescent markers and collecting optical sections, researchers can reconstruct developing neural architecture and follow processes such as cell proliferation, differentiation, migration, and circuit formation over time. In developmental biology, these images reveal how genetic programs and tissue interactions shape visual centers, helping researchers identify developmental defects and compare normal and altered nervous-system development. The approach also supports studies of neural patterning, connectivity, and the formation of functional visual circuits.

Optical-resolution photoacoustic microscopy (OR-PAM) is an emerging technology capable of imaging optical absorption contrasts in vivo with cellular resolution and sensitivity. Here, we provide a visualized instruction on the experimental protocols of OR-PAM, including system configuration, system alignment, typical in vivo experimental procedures, and functional imaging schemes.

Hybridization Chain Reaction FISH in the Developing Drosophila Optic Lobe

This protocol describes the steps required to perform hybridization chain reaction fluorescent in situ hybridization (HCR-FISH) in Drosophila melanogaster larval optic lobes. Procedures for probe design, hybridization, amplification, and tissue mounting are detailed to facilitate visualization of the developing optic lobe.

Optical Materialography Part 2: Image Analysis

2023

Source: Faisal Alamgir, School of Materials Science and Engineering, Georgia Institute of Technology, Atlanta, GA The imaging of microscopic structures of solid materials, and the analysis of the structural components imaged, is known as materialography. Often, we would like to quantify the internal three-dimensional microstructure of a material using only the structural features evidenced by an exposed two-dimensional surface. While X-ray based tomographical methods can reveal buried...
