Atp Dependent Luminescence

ATP-dependent luminescence is a bioluminescence-based method that measures adenosine triphosphate (ATP) through light production, providing a rapid indicator of metabolically active biological material. In a typical assay, luciferase catalyzes the oxidation of luciferin in the presence of ATP, oxygen, and cofactors, and the emitted light is quantified as a signal related to ATP abundance under defined conditions. In immunology and infection research, the method can estimate viable microbial burden or host-cell viability and evaluate antimicrobial or cytotoxic effects. Its speed and sensitivity support screening, process monitoring, and comparisons of cellular responses, although ATP measurements require standardized sampling and interpretation.

The ATP Bioluminescence Assay

2023

In fireflies, the luciferase enzyme converts a compound called luciferin into oxyluciferin, and produces light or “luminescence” as a result. This reaction requires energy derived from ATP in order to proceed, so researchers have exploited the luciferase-luciferin interaction to gauge ATP levels in cells. Given ATP’s role as the cell’s currency of energy, the ATP bioluminescence assay can provide insight into cellular metabolism and overall cell health. In this video, JoVE discusses cellular...

ATP Yield

Cellular respiration produces 30 - 32 ATP per glucose molecule. Although most of the ATP results from oxidative phosphorylation and the electron transport chain (ETC), 4 ATP are gained beforehand (2 from glycolysis and 2 from the citric acid cycle). The ETC is embedded in the inner mitochondrial membrane and is comprised of four main protein complexes and an ATP synthase. NADH and FADH2 pass electrons to these complexes, which pump protons into the intermembrane space. This distribution of...

Phase-Dependent Control of Trap Depth and Persistent Luminescence in Strontium Aluminate Phosphors

2025

This study investigates the standard optical measurements of two persistent luminescent ceramics: SAO-B (blue) and SAO-G (green), such as thermoluminescence (TL) and persistent luminescence (PersL). By controlling electron trap depth, this work innovates 'temperature-resolved' and 'time-resolved' dynamic anti-counterfeiting methods.

Luminescence-Based Assay to Assess Neurotoxicity of Test Compounds on Neurons

2025

In this video, we describe a luminescence-based method to assess the neurotoxicity of neurite outgrowth-promoting small molecule compounds on the viability of hNPC-derived neurons. The assay measures intracellular ATP level, an indicator of cell viability, to evaluate neurotoxicity following test compound treatment.

Here we describe biochemical assays that can be used to characterize ATP-dependent chromatin remodeling enzymes for their abilities to 1) catalyze ATP-dependent nucleosome sliding, 2) engage with nucleosome substrates, and 3) hydrolyze ATP in a nucleosome- or DNA-dependent manner.
