Cd28-mediated Costimulation

CD28-mediated costimulation is a key signaling process that enables full T-cell activation after antigen recognition, helping coordinate adaptive immune responses to infection. When the T-cell receptor binds peptide–MHC complexes, CD28 engages the B7 molecules CD80 or CD86 on antigen-presenting cells, amplifying intracellular signaling and promoting interleukin-2 production, cell survival, proliferation, and differentiation. This second signal helps distinguish productive antigen recognition from incomplete stimulation and supports the formation of effective effector and memory T-cell responses. Understanding CD28 signaling is important for explaining immune defense, immunodeficiency, autoimmunity, and therapies that modulate T-cell activity.

The Upf proteins that carry out nonsense-mediated decay (NMD) are found in all eukaryotic organisms, including humans. Each protein has an individual role, but they need to work in collaboration. Cell-mediated lymphocytotoxicity (CML) assays can be used to test autoreactive responses and study mechanisms of cell death in vitro. However, using live-cell confocal microscopic imaging techniques with fluorescent dyes, the type and kinetics of cell death as well as the pathways utilized can be studied in greater detail.
