Chemical Degranulation

Chemical degranulation is the chemically induced release of preformed granule contents from immune cells, especially mast cells, and is important for understanding rapid inflammatory responses. In mast cells, chemicals such as compound 48/80 or calcium ionophores can raise intracellular calcium levels, activate signaling pathways, and trigger granule fusion with the plasma membrane. Released mediators, including histamine, proteases, and other inflammatory molecules, influence vascular permeability, smooth-muscle activity, and recruitment of immune cells. In immunology and infection research, chemical degranulation assays help evaluate mast-cell activation, characterize inflammatory mechanisms, and assess compounds that may suppress or provoke immune responses.

Mast Cell Degranulation Assay to Study the Effect of a Target Chemical

This video demonstrates a colorimetric assay to determine the degranulation activity of mast cells. The enzyme released by the mast cells hydrolyzes the substrate and produces a colored product. The degranulation activity of the mast cells was measured as the percentage release of enzymes outside the cell normalized with the total enzyme content in the cell.
