Digital Colony Analysis

Digital colony analysis is a computer-assisted method for detecting, counting, and characterizing microbial colonies on culture plates, making growth measurements more consistent and scalable than manual inspection. The process uses digital images and image-analysis algorithms to distinguish colonies from the agar background, segment individual growth areas, and quantify features such as number, size, shape, and pigmentation under defined culture conditions. In immunology and infection research, these measurements support antimicrobial susceptibility testing, pathogen enumeration, host-microbe studies, and evaluation of immune-mediated inhibition. Standardized digital records can improve reproducibility, reduce observer bias, and enable longitudinal comparison of infection-related phenotypes.

ScanLag: High-throughput Quantification of Colony Growth and Lag Time

ScanLag is a high-throughput method for measuring the delay in growth, namely lag time, as well as the growth rate of colonies for thousands of cells in parallel. Screening using ScanLag enables the discrimination between long lag-time and slow growth at the level of a single variant.
