Double Antigen Sandwich

Double antigen sandwich is an immunoassay format that detects antibodies by capturing them between two copies of a specific antigen, making it useful for identifying immune responses to infection. In a typical enzyme-linked immunosorbent assay, one antigen is immobilized on a plate, patient antibodies bind to its epitopes, and an enzyme-labeled antigen attaches to the captured antibodies; after washing and substrate addition, the resulting color signal reflects antibody concentration. This format can measure total antigen-specific antibodies without relying on a secondary anti-immunoglobulin antibody. It supports serological diagnosis, surveillance of exposure, vaccine-response assessment, and evaluation of pathogen-specific immunity.

Snap Chip for Cross-reactivity-free and Spotter-free Multiplexed Sandwich Immunoassays

We demonstrate a snap chip technology for performing cross-reactivity-free multiplexed sandwich immunoassays by simply snapping two slides. A snap apparatus is used for reliably transferring reagents from microarray-to-microarray. The snap chip can be used for any biochemical reactions requiring colocalization of different reagents without cross-contamination.

This video showcases an indirect enzyme-linked immunosorbent assay or ELISA sandwich immunocapture assay designed to assess immunogenic glycoprotein levels in vaccines. The assay plate, containing glycoprotein-specific antibodies, is incubated with serially diluted vaccine samples, including reference and test vaccines, followed by immunoassay to quantify glycoprotein content in the test vaccine.
