Fluorescent Antibody Labeling

Fluorescent antibody labeling is a method that attaches fluorescent dyes to antibodies, enabling specific molecules or cells to be visualized by fluorescence microscopy or flow cytometry. Antibodies are chemically linked to fluorophores, often through reactive groups on the dye and antibody, while preserving the antibody’s ability to recognize its target antigen. In immunology and infection research, labeled antibodies identify pathogens, detect host immune markers, and localize antigens in tissues or cultured cells. The resulting signal supports diagnostic assays, phenotyping of immune cells, and analysis of infection processes, although labeling conditions and fluorophore selection must be optimized to limit background and maintain specificity.

This protocol describes single-antibody labeling (SAL) to resolve the nanoscale spatial organization of plasma membrane proteins. By leveraging cumulative antibody-epitope interactions at the single-molecule level, membrane SAL (mSAL) maps local epitope distributions while simultaneously capturing antibody binding behavior in the native cellular environment.
