Real Time Cytotoxicity

Real-time cytotoxicity is the continuous measurement of target-cell injury or death as it occurs, providing a dynamic view of immune and infection-related damage rather than a single endpoint. Assays monitor changes such as cell adherence, morphology, membrane integrity, or viability signals over time, allowing cytotoxic responses to be quantified as they develop. In immunology, this approach can assess how cytotoxic T cells, natural killer cells, antibodies, or other effectors eliminate infected or abnormal cells. Time-resolved measurements help reveal response kinetics, compare treatment conditions, and distinguish rapid killing from delayed or progressive cytotoxic effects.

Autonomously Bioluminescent Mammalian Cells for Continuous and Real-time Monitoring of Cytotoxicity

Mammalian cells expressing the bacterial bioluminescence gene cassette (lux) produce light autonomously. The resulting bioluminescent dynamics upon chemical exposure have been demonstrated to reflect the treatment effects on cellular growth and metabolism, making these cells an inexpensive, continuous, real-time toxicity screening tool that can easily be adapted for high-throughput automation.

High Throughput, Real-time, Dual-readout Testing of Intracellular Antimicrobial Activity and Eukaryotic Cell Cytotoxicity

A high throughput, real-time assay was developed to simultaneously identify (1) eukaryotic cell-penetrant antimicrobials targeting an intracellular bacterial pathogen, and (2) assess eukaryotic cell cytotoxicity. A variation on the same technology was thereafter combined with digital dispensing technology to enable facile, high-resolution, dose-response, and two- and three-dimensional synergy studies.

The whole blood cytotoxicity assay (WCA) is a cytotoxicity assay developed by incorporating high-throughput cell positioning technology with fluorescence microscopy and automated image processing. Here, we describe how lymphoma cells treated with an anti-CD20 antibody can be analyzed real-time in human whole blood to provide quantitative cellular cytotoxicity analysis.
