Single Nucleotide Polymorphism Analysis

Single nucleotide polymorphism (SNP) analysis is the detection and interpretation of single-base differences in DNA, which can reveal genetic variation among individuals or populations. The process compares targeted sequences using methods such as PCR, allele-specific probes, genotyping arrays, or DNA sequencing to identify which nucleotide is present at a defined genomic position. In immunology and infection research, SNP analysis helps examine how host variants influence immune responses, susceptibility to pathogens, disease severity, or treatment outcomes, while pathogen SNP profiles can support strain tracking and transmission studies. These findings strengthen genetic association studies and inform personalized approaches to infectious disease research.

This work presents a simple and visual method to detect multi-nucleotide polymorphisms on a pneumatic droplet manipulation platform. With the proposed method, the entire experiment, including droplet manipulation and detection of multi-nucleotide polymorphisms, can be performed near 23 °C without the aid of advanced instruments.

While high resolution melting analysis offers the ability to differentiate between single nucleotide polymorphisms in a heterogeneous population, mutant allele amplification bias can increase its ability to detect alleles present at relatively low percentages within a sample.
