Split-thickness Skin

Split-thickness skin is a skin graft containing the epidermis and part of the dermis, used to cover wounds when tissue loss prevents normal closure. During harvesting, a thin layer is removed from a donor site and transferred to the wound, where it revascularizes and supports restoration of the protective barrier; the donor site can regenerate epidermis from remaining skin structures. In immunology and infection research, split-thickness grafts help investigate host defenses, inflammation, microbial colonization, and tissue repair after injury. Clinically, they provide wound coverage and reduce fluid loss, while experimental models clarify how immune responses influence graft integration and infection risk.

The goal of this protocol is to build up a three-dimensional full thickness skin equivalent, which resembles natural skin. With a specifically constructed automated wounding device, precise and reproducible wounds can be generated under maintenance of sterility.
