Viral Neuroinflammatory Response

Viral neuroinflammatory response is the coordinated immune reaction that occurs when a viral infection affects the nervous system, helping control infection but potentially injuring neural tissue. Viral detection by innate immune receptors activates microglia, astrocytes, and infiltrating leukocytes, which release cytokines and chemokines that recruit additional immune cells and shape adaptive antiviral responses. This process can disrupt the blood-brain barrier, alter neuronal function, and contribute to encephalitis or longer-term neurological damage when inflammation is excessive or unresolved. Studying these mechanisms supports research into viral pathogenesis, biomarkers, therapeutic targets, and strategies that balance antiviral defense with protection of the brain.

The footpad inoculation model is a valuable tool for characterizing viral-induced neuroinflammatory responses in vivo. In particular, it provides a clear assessment of viral kinetics and associated immunopathological processes initiated in the peripheral nervous system.

Levels of the inflammatory cell-signaling molecule nitric oxide (NO) are commonly assayed using Griess reagent. In this protocol, we have created a modified Griess assay utilizing live Drosophila brain tissue in order to detect the secretion of NO in a simple, quantifiable and highly repeatable method.
