Chemoresistant Cscs

Chemoresistant CSCs are cancer stem cells that survive anticancer drug treatment and can regenerate tumor cell populations, making them important in cancer persistence and relapse. Their resistance commonly reflects a combination of stem-like self-renewal, cellular quiescence, enhanced DNA-damage repair, drug-efflux activity, and reduced susceptibility to apoptosis, although the dominant mechanisms vary by tumor type. In medicine, studying these cells helps explain incomplete treatment responses and informs strategies that combine conventional chemotherapy with therapies targeting stem-cell maintenance or resistance pathways. Measuring CSC frequency and treatment response can also support biomarker development and more durable therapeutic design.

Cancer stem cells (CSCs) are implicated in tumor relapse due to chemoresistance. We have optimized a protocol for selection and expansion of CSCs from ovarian cancer cell lines. By treating cells with the chemotherapeutic cisplatin and culturing in a stem cell promoting media we enrich for non-adherent CSC cultures.
