Diabetic Neuropathy

Diabetic neuropathy is a group of nerve disorders caused by diabetes, most commonly affecting the peripheral nerves in the feet and legs and sometimes the hands and internal organs. Persistently elevated blood glucose disrupts nerve metabolism and damages the small blood vessels that supply nerves, leading to impaired signaling, inflammation, and progressive nerve injury. Symptoms may include burning pain, tingling, numbness, muscle weakness, or loss of protective sensation, which increases the risk of unnoticed wounds and infection. Understanding diabetic neuropathy supports early screening, glucose management, symptom control, and strategies to prevent disability and other complications.

DefinitionDiabetic neuropathy is nerve damage caused by long-standing diabetes mellitus. It results directly from prolonged high blood sugar levels.PathophysiologyThe pathophysiology of diabetic neuropathy involves both metabolic and vascular disturbances triggered by chronic hyperglycemia.Metabolic injury: Elevated glucose levels activate the polyol pathway within nerve cells, leading to the accumulation of sorbitol and fructose. This increases oxidative stress, disrupts normal nerve...

This video demonstrates a method for establishing a small fiber neuropathy model in mice using the TRPV1 agonist resiniferatoxin (RTX). It outlines the steps involved in administering RTX to mice and explains how RTX induces neuronal death in small-diameter sensory neurons, thereby establishing the neuropathy model.

The goal of this protocol is to photochemically induce ischemic injury to the posterior optic nerve in rat. This model is critical to studies of the pathophysiology of posterior ischemic optic neuropathy, and therapeutic approaches for this and other optic neuropathies, as well as of other CNS ischemic diseases.
