Enhanced Permeability Retention

Enhanced Permeability Retention is a biological phenomenon in which macromolecules and nanoparticles accumulate preferentially in tumor tissue because abnormal blood vessels allow them to enter while poor lymphatic drainage limits their removal. Rapidly growing tumors often develop irregular, leaky vasculature with widened endothelial gaps, creating increased permeability compared with healthy tissue. In medicine, this effect supports passive tumor targeting for nanocarrier-based drug delivery, including liposomes and polymeric nanoparticles, with the goal of concentrating therapeutics at diseased sites and reducing systemic exposure. However, tumor heterogeneity, vascular structure, and differences in circulation can limit its consistency, making the EPR effect an important focus of drug-delivery research.
