Estrous Cycle

The estrous cycle is the recurring sequence of reproductive, hormonal, and behavioral changes that prepares many female mammals for ovulation, mating, and possible pregnancy. It is regulated by communication among the hypothalamus, pituitary gland, and ovaries: gonadotropin-releasing hormone stimulates follicle-stimulating and luteinizing hormone release, while changing estrogen and progesterone levels control follicle development, ovulation, uterine preparation, and receptivity. Understanding the estrous cycle supports veterinary fertility management, breeding decisions, and interpretation of reproductive disorders. It also provides a framework for using animal models to study endocrine regulation, contraception, pregnancy, and reproductive medicine.

Here, we describe how to identify the stage of the murine reproductive (proestrus, estrus, metestrus, or diestrus) by simple, non-invasive collection and cytological assessment of vaginal smear samples. We further describe how vaginal cytology reflects circulating hormonal levels underlying transition through the murine reproductive cycle.
