Phenotypic Resistance Confirmation

Phenotypic resistance confirmation is the laboratory process of verifying that a microorganism can withstand an antimicrobial drug under defined testing conditions, providing evidence that resistance affects actual growth or survival. In antimicrobial susceptibility testing, a clinical isolate is exposed to selected agents at standardized concentrations, and growth is assessed using measures such as inhibition zones or minimum inhibitory concentrations; results are interpreted against established breakpoints. This confirmation supports accurate treatment selection, helps distinguish clinically meaningful resistance from suspected resistance based on genetic markers, and strengthens surveillance of emerging drug-resistant pathogens. In medicine, it can guide infection management and inform infection-control decisions.

The confirmation bias is the tendency to focus on information that confirms our existing beliefs and ignore information that is inconsistent with our expectations. For example, if you think that your professor is not very nice, you notice all of the instances of rude behavior exhibited by the professor while ignoring the countless pleasant interactions he is involved in on a daily basis. Have you ever fallen prey to the confirmation bias, either as the source or target of such bias? Another...

2015

Drug resistance to targeted therapeutics is widespread and the need to identify mechanisms of resistance--prior to or following clinical onset--is critical for guiding alternative clinical management strategies. Here, we present a protocol to couple derivation of drug-resistant lines in vitro with sequencing to expedite discovery of these mechanisms.

Humane Euthanasia and Confirmation of Death in Rodents

All animal procedures described here must be conducted in accordance with institutional animal ethics guidelines and approved by IACUC. All procedures must follow the principles of the 3Rs—Replacement, Reduction, and Refinement—and must be performed by trained personnel. Humane endpoints in rodents are predefined clinical, behavioral, or physiological indicators that signal an animal is experiencing significant pain, distress, or declining health. When these signs are observed, timely...
