Targeted Therapy Resistance

Targeted therapy resistance is the loss of cancer responsiveness to a treatment designed to inhibit a specific molecular target, limiting the therapy’s clinical benefit. Resistance can develop through secondary mutations that alter the drug-binding site, activation of bypass signaling pathways, changes in cancer-cell identity, or reduced drug exposure from altered transport and metabolism. In medicine, identifying these mechanisms through tumor profiling, repeat biopsy, or liquid biopsy can guide treatment selection and the design of combination or sequential therapies. Studying resistance also supports biomarker development, treatment monitoring, and clinical trials aimed at extending durable responses.

The targeted cancer therapies, also known as “molecular targeted therapies,” take advantage of the molecular and genetic differences between the cancer cells and the normal cells. It needs a thorough understanding of the cancer cells to develop drugs that can target specific molecular aspects that drive the growth, progression, and spread of cancer cells without affecting the growth and survival of other normal cells in the body. There are several types of targeted therapies against specific...
