Therapeutic Treatment Efficacy

Therapeutic treatment efficacy is the extent to which a medical intervention produces its intended health benefit under defined conditions, making it central to judging whether a therapy works. It is assessed by comparing prespecified clinical or biological outcomes in treated patients with those in an appropriate comparator, while accounting for dose, treatment duration, patient characteristics, and adverse effects. In medicine, efficacy evidence supports clinical decision-making, regulatory evaluation, treatment selection, and optimization of care. Distinguishing efficacy from effectiveness, which reflects performance in routine practice, helps researchers interpret trial findings and determine how reliably results may translate to broader patient populations.

We describe methods for longitudinal monitoring of the efficacy of therapeutics for the treatment of colonic pathologies in mice using a rigid endoscope. This protocol can be readily used for the characterization of the therapeutic response of an individual tumor in live mice and also for monitoring potential disease relapse.

Bioimaging methods used to assess cell biodistribution of nanoparticles are applicable for therapeutic and diagnostic monitoring of nanoformulated compounds. The methods described herein are sensitive and specific when assessed by histological coregistration. The methodologies provide a translational pathway from rodent to human applications.
