Simple Western

Simple Western is an automated protein analysis method that separates proteins by size in a capillary and detects them with antibody-based chemiluminescence or fluorescence. After electrophoretic separation, proteins are immobilized to the capillary wall, exposed to primary and labeled secondary antibodies, and quantified from signal intensity relative to a protein standard or loading control. In neuroscience, this workflow measures neuronal proteins, phosphorylation states, receptors, and signaling molecules from small or limited samples, supporting studies of synaptic plasticity, neurodegeneration, and neural development. Automation reduces hands-on processing and sample consumption while improving consistency compared with conventional Western blotting.

This protocol explores the latest advancements in performing Western blot analyses. These novel modifications employ a Bis-Tris gel system with a 35 min electrophoresis run time, a 7 min dry blotting transfer system, and infrared fluorescent protein detection and imaging that generates higher resolution, quality, sensitivity, and improved accuracy of Western data.
