Tangential Sectioning

Tangential sectioning is a histological method that cuts tissue parallel to its surface, providing a broad view of structures arranged across a tissue plane. In neuroscience, the specimen is carefully oriented before serial sections are collected, allowing cortical layers, neuronal processes, and fiber pathways to be examined along their horizontal organization rather than primarily through tissue depth. This approach supports microscopic analysis of cortical maps, dendritic and axonal patterns, and regional connectivity, especially when combined with staining or immunohistochemistry. By preserving spatial relationships across the tissue surface, tangential sections help researchers investigate neural architecture, developmental patterning, and changes associated with neurological disease.

Tangential flow ultrafiltration (TFU) is a recirculation method used for the weight-based separation of biosamples. TFU was adapted to size-select (1-20 nm diameter) and highly concentrate a large volume of polydisperse silver nanoparticles (4 L of 15.2 μg ml-1 down to 4 ml of 8,539.9 μg ml-1) with minimal aggregation.

Preparation of Adult Drosophila Eyes for Thin Sectioning and Microscopic Analysis

A standard approach to prepare adult Drosophila eyes for semi-thin sectioning and light microscopic analysis is presented here. The protocol can be used for gross morphological analysis of eye defects, or with the indicated adjustments can be used to determine genetic requirements of genes in specific cell types of the eye (e.g. clonal analysis of photoreceptors) or for electron microscopic analysis.
