Pleiotropy means that a single gene affects multiple distinct traits in the body.
For example, in humans, the SOX10 gene codes for a transcription factor that is important for embryonic development and affects multiple traits.
Genetic mutations in the SOX10 gene, such as deletions, can produce a nonfunctional protein that disrupts normal pigment formation in the body.
This leads to features such as white hair patches, lighter skin areas, and irises with different colors.
A mutation in the SOX10 gene can also disrupt the development of structures in the inner ear, leading to hearing loss or deafness.
A mutation in the SOX10 gene can also disrupt nerve development in the large intestine.
As a result, the intestinal wall cannot move contents forward properly.
This causes buildup in the intestine, which can lead to blockage and enlargement of the colon over time.
Clinically, this appears as abdominal distension, discomfort or pain, vomiting, and severe constipation.
These combined effects on pigmentation, hearing, and intestinal function are characteristic of Waardenburg syndrome type 4, or WS4.
